Lysosome targeting fluorescent probe for NAAA imaging and its applications in the drug development for anti-inflammatory
Laboratory studyOther animals
- Design
- Laboratory study
- Subjects
- Living cells including RAW264.7 mouse macrophages; 42 traditional Chinese medicines screened
- Dose used in the study
- Not stated in the abstract; palmitoylethanolamide was not given as a treatment
- Duration
- Not stated in the abstract
- What was measured
- Performance of a new fluorescent probe (PMBD) for imaging NAAA, the enzyme that breaks down palmitoylethanolamide, and screening of NAAA inhibitors
What the authors reported
The study found:
- The probe detected endogenous NAAA selectively in living cells and localised to lysosomes.
- A high-throughput NAAA inhibitor assay built on the probe screened 42 anti-inflammatory traditional Chinese medicines.
- Ellagic acid from Cornus officinalis was identified as an NAAA inhibitor and suppressed LPS-induced iNOS and nitric oxide in RAW264.7 cells.
Limits of this study
A chemistry and cell study; it cannot show effect in people or animals. PEA itself was not tested; the work concerns the enzyme that degrades it. The authors declare no competing financial interests.
