The Effects of Palmitoylethanolamide or Ibuprofen on the Abundance Profile and Synthesis Rate of Proteins in C2C12 Skeletal Myotubes

Paige L Cole, Connor A Stead, Jatin G Burniston, Daniel J Owens. FASEB Bioadv. 2026 Apr 23;8(4):e70108.

Laboratory studyOther animals

Design
Laboratory study
Subjects
Differentiated C2C12 mouse skeletal myotubes (cell culture)
Dose used in the study
PEA 10 micromolar or ibuprofen 100 micromolar, treated for 36 hours
Duration
36 hours
What was measured
Protein synthesis rates (1541 proteins) and relative protein abundance (3085 proteins) using D2O labelling, comparing PEA and ibuprofen with vehicle control.

What the authors reported

PEA increased synthesis of 101 proteins and reduced synthesis of 2, mostly ribosomal proteins; ibuprofen increased synthesis of 165 proteins and reduced 7, with broader changes across muscle contraction, extracellular matrix, carbohydrate and lipid metabolism proteins. Both increased total ribosomal protein synthesis by about 80% and 40S ribosomal subunit abundance by about 18% relative to control.

Limits of this study

A cell culture study in mouse myotubes; it cannot show effect in muscle in a living animal or person. The authors declare no conflicts of interest.
