NAPE-specific phospholipase D regulates LRRK2 association with neuronal membranes
Laboratory studyHumansOther animals
- Design
- Laboratory study
- Subjects
- Human SH-SY5Y neuroblastoma cells with NAPE-PLD silenced by siRNA; mice lacking NAPE-PLD
- Dose used in the study
- Not applicable; palmitoylethanolamide was not given
- Duration
- Not stated in the abstract
- What was measured
- NAPE levels and the membrane versus cytosol distribution and total content of LRRK2, a kinase linked to Parkinson's disease, after NAPE-PLD deletion or after lowering NAPEs with bacterial PLD
What the authors reported
Loss of NAPE-PLD, the enzyme that makes N-acylethanolamines such as palmitoylethanolamide from NAPEs, caused NAPEs to accumulate, shifted LRRK2 from membranes to cytosol and reduced total LRRK2. Lowering NAPEs with bacterial PLD increased LRRK2 membrane association.
Limits of this study
A cell and mouse study of a lipid pathway; it cannot show any effect in people. PEA itself was not tested. Funding and conflicts not stated in the abstract.
