Endocannabinoids and PEA appear to reduce trigeminal pain signalling through several migraine-related pathways in animal models, and inhibiting endocannabinoid-degrading enzymes has been a research focus, the authors report. They conclude that controlled clinical studies of cannabis are lacking and that ECS-related molecules are potential targets needing further study.
Limits of this study
A narrative review, not a systematic one, and much of the cited evidence is from animal models. It gives no trial numbers for palmitoylethanolamide. Funding and conflicts not stated in the abstract.
