Effects of palmitoylethanolamide on immunologically induced histamine, PGD2 and TNF-alpha release from canine skin mast cells

Cerrato S, Brazis P, della Valle MF, Miolo A, Puigdemont A. Vet Immunol Immunopathol. 2010;133(1):9-15.

Laboratory studyDogs

Design
Laboratory study
Subjects
Mast cells freshly isolated from canine skin biopsies, sensitised with IgE-rich serum and challenged with anti-canine IgE
Dose used in the study
PEA from 10 nanomolar to 10 micromolar in vitro
Duration
In vitro challenge
What was measured
Histamine, prostaglandin D2 and TNF-alpha release

What the authors reported

PEA inhibited release of all three mediators. Peak histamine inhibition was 54 percent at 3 micromolar; PGD2 inhibition 26 percent at 10 micromolar; TNF-alpha inhibition 29 percent at 10 micromolar.

Limits of this study

In vitro cell study. Two authors are from Innovet, which makes veterinary PEA products. Partial inhibition, not abolition, of mediator release.
