Ultramicronized palmitoylethanolamide counteracts the effects of compound 48/80 in a canine skin organ culture model
Laboratory studyDogs
- Design
- Laboratory study
- Subjects
- Skin biopsies from three healthy donor dogs, cultured in triplicate (ex vivo)
- Dose used in the study
- Ultramicronised PEA 30 micromolar in the culture medium, given 24 hours before and 72 hours with compound 48/80 (10 or 100 micrograms/mL), a mast cell secretagogue
- Duration
- 5 days of culture
- What was measured
- Degranulating mast cells per square millimetre; histamine in the medium; capillary diameter; epidermal thickness, keratinocyte proliferation and differentiation markers
What the authors reported
Compound 48/80 caused mast cell degranulation and capillary dilation. PEA reduced degranulating mast cells (p < 0.005 and p < 0.01 at the two concentrations) to levels no different from untreated skin, and reversed the vasodilation at the higher concentration (p < 0.001). Histamine fell by about half but not significantly. PEA alone did not change the epidermis.
Limits of this study
Ex vivo skin from three dogs; the biopsy was the statistical unit. Single time point. Partly funded by Innovet, which makes the PEA product; one author is an Innovet consultant.
