The Comparative Efficacy of Palmitoylethanolamide (PEA) With the Combination of Pregabalin and Nortriptyline on Post-extraction Trigeminal Neuropathy by Using Magnetic Resonance (MR) Neurography: A Randomized Clinical Trial
Randomised controlled trialHumansWith pregabalin and nortriptyline (a separate comparator arm, and also combined with palmitoylethanolamide in a third arm)
This study tested palmitoylethanolamide together with pregabalin and nortriptyline (a separate comparator arm, and also combined with palmitoylethanolamide in a third arm). Its results cannot be assigned to palmitoylethanolamide alone. More combination studies.
- Design
- Randomised controlled trial
- Subjects
- 60 patients with post-extraction trigeminal neuropathy (20 per group, three groups)
- Dose used in the study
- Group I: pregabalin 75 mg plus nortriptyline 10 mg once daily; Group II: palmitoylethanolamide 600 mg twice daily; Group III: the combination of both
- Duration
- 12 weeks
- What was measured
- Pain intensity on a numeric rating scale (primary outcome) and nerve signal intensity and thickness on magnetic resonance neurography (secondary outcome)
What the authors reported
All three groups had a significant decrease in mean pain scores after treatment, with the combination group showing the greatest reduction. Palmitoylethanolamide significantly reduced pain scores with negligible side effects.
Limits of this study
A randomised trial in post-extraction trigeminal neuropathy only; results may not extend to other neuropathic pain conditions. Funding and conflicts not stated in the abstract beyond a general declaration of no competing interests.
