Intracellular Ca2+-dependent formation of N-acyl-phosphatidylethanolamines by human cytosolic phospholipase A2ε
Laboratory studyHumans
- Design
- Laboratory study
- Subjects
- A human embryonic kidney 293 cell line engineered so doxycycline induces cPLA2-epsilon
- Dose used in the study
- No palmitoylethanolamide was given; cells were treated with doxycycline and a calcium ionophore
- Duration
- Not stated in the abstract
- What was measured
- Endogenous substrates and products of cPLA2-epsilon by liquid chromatography-tandem mass spectrometry, including N-acyl-phosphatidylethanolamines (precursors of palmitoylethanolamide) and N-acylethanolamines
What the authors reported
Induced cells given a calcium ionophore produced large amounts of diacyl and alkenylacyl NAPEs and of NAEs, while diacyl and alkenylacyl PEs and diacyl PCs fell and lysoPEs and lysoPCs rose. cPLA2-epsilon appears to make NAPEs in a calcium-dependent way, using PEs as acyl acceptors and PCs and PEs as donors.
Limits of this study
A cell-line biochemistry study of how PEA precursors are made; it cannot show effect in people or animals. Funding and conflicts not stated in the abstract.
