Synergistic modulation of nociceptive and inflammatory pain by palmitoylethanolamide and Cucumis sativus extract in a monosodium-iodoacetate induced osteoarthritic rat model

Om Prakash Sharma, Radika Sharma. Inflammopharmacology. 2026 May;34(5):3335-3345.

Laboratory studyOther animalsWith Cucumis sativus extract (branded Ostizeel)

This study tested palmitoylethanolamide together with Cucumis sativus extract (branded Ostizeel). Its results cannot be assigned to palmitoylethanolamide alone. More combination studies.

Design
Laboratory study
Subjects
42 female Wistar rats with monosodium iodoacetate-induced osteoarthritis, in seven groups
Dose used in the study
PEA, Cucumis sativus extract, an NSAID standard, or combinations, given orally for 28 days
Duration
28 days
What was measured
Nociceptive behaviour (movement-evoked pain, Randall-Selitto, von Frey, weight-bearing), serum interleukin-1beta and MMP-13, and drug interaction using Bliss Independence and combination index analysis

What the authors reported

The PEA plus Cucumis sativus combination improved pain thresholds and limb function more than either alone (p<0.0001) and cut serum IL-1beta and MMP-13 by about 50%. Bliss Independence analysis showed positive synergy scores across behavioural and biochemical measures.

Limits of this study

A rat model of chemically induced osteoarthritis; it cannot show effect in people or animals with naturally occurring osteoarthritis. The authors declare no competing interests; the study used a proprietary combination (Ostizeel) from a commercial research organisation.
