Laboratory studyOther animalsWith ARN19702, ARN077 and ARN726 (NAAA inhibitors that raise endogenous PEA and OEA, tested alone)

This study tested palmitoylethanolamide together with ARN19702, ARN077 and ARN726 (NAAA inhibitors that raise endogenous PEA and OEA, tested alone). Its results cannot be assigned to palmitoylethanolamide alone. More combination studies.

Design
Laboratory study
Subjects
Marchigian Sardinian alcohol-preferring rats (numbers not stated in the abstract)
Dose used in the study
ARN19702 3 and 10 mg/kg intraperitoneally; ARN19702 1, 3 and 10 microgram/microlitre, ARN077 and ARN726 3 and 10 microgram/microlitre by microinjection into the ventral tegmental area or nucleus accumbens
Duration
Not stated in the abstract
What was measured
Voluntary alcohol intake in a two-bottle choice test; operant alcohol self-administration under fixed-ratio and progressive-ratio schedules; water, food and saccharin intake.

What the authors reported

This laboratory study found:

  • Intraperitoneal ARN19702 lowered voluntary alcohol intake dose-dependently, about 47% at 10 mg/kg (p<0.001), without affecting water, food or saccharin.
  • It also reduced alcohol self-administration under both schedules.
  • Microinjection of all three NAAA inhibitors into the ventral tegmental area reduced self-administration, but injection into the nucleus accumbens did not.

Limits of this study

A rat study; it cannot show effect in people. PEA itself was not given. Daniele Piomelli is an inventor on University of California patent applications describing NAAA inhibitors.
