Ultra-micronized Palmitoylethanolamide Effects on Sleep-wake Rhythm and Neuropathic Pain Phenotypes in Patients with Carpal Tunnel Syndrome: An Open-label, Randomized Controlled Study

Evangelista M, Cilli V, De Vitis R, Militerno A, Fanfani F. CNS & Neurological Disorders - Drug Targets, 2018; 17(4):291-298.

Controlled trialHumans

Design
Controlled trial
Subjects
42 patients awaiting carpal tunnel surgery, with sleep disturbance and pain, randomised to two groups
Dose used in the study
600 mg of ultramicronised palmitoylethanolamide twice a day, before and after surgery. The comparison group had surgery only.
Duration
The period before and after surgery
What was measured
Sleep quality on the Pittsburgh Sleep Quality Index as the main outcome, and pain on a numeric rating scale

What the authors reported

Sleep quality and pain both improved significantly in the treated group by the end of the pre-surgery period (p < 0.0001 for each), results the authors describe as preliminary.

Limits of this study

Open-label, with no placebo. The comparison group received nothing, so patients knew whether they were treated, and both outcomes were self-reported. The blinded trial by Faig-Martí 2017 in the same condition found no advantage over placebo. The entry was checked against the abstract only. Funding was not listed.
