Palmitoylethanolamide and hemp oil extract exert synergistic anti-nociceptive effects in mouse models of acute and chronic pain
Laboratory studyOther animalsWith hemp oil extract
This study tested palmitoylethanolamide together with hemp oil extract. Its results cannot be assigned to palmitoylethanolamide alone. More combination studies.
- Design
- Laboratory study
- Subjects
- Mice tested in the formalin (acute pain) and chronic constriction injury (CCI, chronic pain) models
- Dose used in the study
- Hemp oil extract (HOE) 10, 50 or 100 mg/kg oral, alone or combined with PEA 10, 30, 100 or 300 mg/kg oral
- Duration
- CCI effects assessed to day 7 after surgery
- What was measured
- Pain-related behaviours in both models; IL-6 and IL-10 transcription in lumbar spinal cord after CCI; pharmacokinetics of PEA with and without HOE
What the authors reported
The study found:
- HOE alone gave modest pain relief, but sub-effective doses of HOE combined with PEA gave a greater-than-additive reduction in pain behaviours.
- Both HOE alone and the HOE/PEA combination reduced the rise in spinal IL-6 and IL-10 after CCI to a similar extent.
- HOE co-administration raised and prolonged PEA blood levels.
Limits of this study
A mouse-model study; it cannot show effect in people. Funding and conflicts not stated in the abstract.
